The 25-year-old's one minute 46.44 seconds was bettered only by the Kenyan, who eased clear down the home straight to cross in 1:45.14 at New York's Icahn Stadium.
Osagie had run more than a second faster at the last event in Doha two weeks ago but held off the likes of Olympic bronze medallist Timothy Kitum in wet and windy conditions to provide a rare British highlight.
Shara Proctor was second in the long jump with a leap of 6.72m, 0.07m behind winner Jenay Deloach Soukup, the 2012 Olympic bronze medallist, who set a meet record.
But Christine Ohuruogu had a tougher time, struggling to sixth in a 400m race that Botswana's Amantle Montsho won with comfort in 49.91 seconds.
Sanya Richards-Ross had been expected to feature for the first time since winning Olympic 400m gold in London but was a no-show at the start line, explaining on Twitter that she had not yet recovered from a toe operation.
European Indoor pole vault champion Holly Bleasdale pulled out of the competition after the first round, where she finally cleared her opening height of 4.23m at the third attempt, while Phillips Idowu did not appear in the triple jump, which was won by Frenchman Benjamin Compaore.
Elsewhere, Tyson Gay roared to a 100m victory in 10.02 seconds, beating compatriot Ryan Bailey (10.15s) and Keston Bledman of Trinidad and Tobago (10.16s).
Gay, a former 100m and 200m world champion who won the Jamaica Invitational earlier this month in a world-leading 9.86 seconds, is pondering a 100m-200m world double bid if he can qualify at next month's US nationals.
With August's World Championships in Moscow drawing ever closer, the Diamond League meetings in Birmingham on 30 June and London on 26-27 July are set to draw a star cast.
