It was the first time in seven attempts John Coleman's men had beaten their neighbours since returning to the Football League and ended a run of five successive defeats against Keith Hill's team.
They took the lead when James Ryan's free-kick from the left was flicked on by Michael Symes and headed home by Robert Grant.
Symes wrapped up the points in the 69th minute when he collected a long punt forward from Phil Edwards before volleying home a stunning 30-yard effort.
Chris Dagnall pulled one back in the 91st minute for Rochdale, but Accrington held on for the win.
